Abstract
During her Ph.d. studies, Marlene Andersen investigated the regulation of pro-resolving, anti-inflammatory biologic systems in rheumatoid arthritis. Marlene Andersen has shown that important pro-resolving and anti-inflammatory mechanisms are regulated in pathogenetically important leukocyte subsets in rheumatoid arthritis. Through measurements of the reaction of the gene expression of resistin and melanocortin receptors (MCRs) to treatment with TNFα inhibitor adalimumab it was found that gene expressions decreased significantly in pathogenic leukocyte subsets. The decreases were related to fall in the gene expressions of important disease driving cytokines, such as interferon γ and interleukin 1 β in T cytotoxic- and T helper cells, respectively. Changes in resistin and transforming growth factor β gene expressions in monocytes correlated.
The achieved scientific results, i.e. the finding of a regulated melanocortin system in T cytotoxic cells and B-cells as well as a relation between MCR and Th1 cytokines, uncover, together with signs of a regulatory role for resistin, new pathways, which may serve as paths to the induction of curative immune tolerance in rheumatoid arthritis.
